Abstract

The prostate gland can experience disorders such as Benign Prostatic Hyperplasia (BPH). BPH is one type of prostate disease characterized by a benign tumor that occurs in males, and its incidence is related to increasing age. Histopathological examination is a method used to detect prostate cancer. Fixation is the initial stage in tissue processing, aiming to prevent autolysis and decay. This study aims to compare the histopathological images of prostate tissue with different fixation times. This quantitative research used a True experimental approach with a Post-test Only Control Group design. The study was conducted in July 2023. Samples were taken using a random sampling technique, comprising 28 preparations with four treatments: fixation for 2 hours, 24 hours, 48 hours, and 72 hours. Data were analyzed using the Pearson chi-square test. The results showed differences in histopathological preparations based on the clarity of the cytoplasm and nucleus of the cells, which were more evident in the 24-hour fixation. Prostate tissue preparations fixed for 2 hours exhibited less distinct nuclei and cytoplasm, and the cells appeared denser. Prostate tissue fixed for 48 hours showed clear cytoplasm and nuclei but slight shrinkage. Prostate tissue fixed for 72 hours showed smaller nuclei and experienced shrinkage. The Pearson Chi-Square analysis indicated significant differences in histopathological preparations based on different fixation times regarding the cytoplasm and nucleus of prostate tissue, with a value of 0.00 (p 0.05).
